چکیده

In previous studies (Tanrhis and Frisch, 1951a,b) it was found that a human blood medium was satisfactory for determining the susceptibility of Mycobacterium tuberculosis to streptomycin. This investigation was undertaken to determine whether the same medium would be suitable also for performing isoniazid susceptibility tests, and to compare it with the American Trudeau Society (Report of the Committee on Evaluation of Laboratory Procedures, 1946) and Herrold (1931) media.

The purification and properties of peroxidase in Mycobacterium tuberculosis H37Rv and its possible role in the mechanism of action of isonicotinic acid hydrazide.

Peroxidase from Mycobacterium tuberculosis H37Rv was purified to homogeneity. The homogeneous protein exhibits catalase and Y (Youatt's)-enzyme activities in addition to peroxidase activity. Further confirmation that the three activities are due to a single enzyme was accomplished by other criteria, such as differential thermal inactivation, sensitivity to different inhibitors, and co-purificat...

The molecular basis of isoniazid resistance in Mycobacterium tuberculosis.

Isoniazid, the hydrazide of isonicotinic acid, was synthesized for the first time in 1912 by two Czech scientists, but its powerful antituberculous activity was detected only in 1951, and the discovery of isoniazid was a milestone in the chemotherapy of tuberculosis. Its activity is excellent against species of the Mycobacterium tuberculosis complex (M. tuberculosis, Mycobacterium bovis, Mycoba...

Synthesis, antitubercular activity and pharmacokinetic studies of some Schiff bases derived from 1-alkylisatin and isonicotinic acid hydrazide (INH).

N'-(1-alkyl-2,3-dihydro-2-oxo-1H-3-indolyliden)-4-pyridinecarboxylic acid hydrazide derivatives, 3(a-g), were synthesized in a trial to overcome the resistance developed with the therapeutic uses of isoniazid (INH). The lipophilicity of the synthesized derivatives supersedes that of the INH as expressed by Clog p values. The synthesized compounds and INH were tested against bovin, human sensiti...

Prevalence ofMultidrug Resistant Mycobacterium tuberculosis by Mycobacteria growth indicator tube in Golestan province, North of Iran

Abstract Background and objectives: Identification and monitoring of multidrug-resistant Mycobacterium tuberculosis strains (MDR) is highlighted by the high risk of their spreading in different areas. Prevalence of these strains was evaluated in Golestan province in northeast of Iran. Material and Methods: Drug susceptibility testing to Isoniazid and rifampin was carried out for 148 clinical sa...

Nitric oxide generated from isoniazid activation by KatG: source of nitric oxide and activity against Mycobacterium tuberculosis.

Isonicotinic acid hydrazide (INH) is a frontline antituberculosis agent. Once taken up by Mycobacterium tuberculosis, INH requires activation by the catalase-peroxidase KatG, converting INH from its prodrug form into a range of bactericidal reactive species. Here we used 15N-labeled INH together with electron paramagnetic resonance spin trapping techniques to demonstrate that nitric oxide (NO*)...
